The mass spectrometry core facility at OUHSC provides services that include protein identification and protein quantification from a wide variety of complexÂ sample types, from cell lysate, protein complexes and plasma.Â
Our principal approach for protein analysis is the "bottom-up" analysis, where the protein samples are proteolytically digested, producing peptides that are signatures of the original proteins.
Protein quantification can be accomplished using a 'gel free, label-free' technology that provides both relative quantification (test vs. control) and absolute quantification (nanograms protein)Â
We currently can performÂ open (unbiased) qualitative and quantitative analyses using high resolution, accurate mass data for high confidence identifications and quantitative reproducibility. This is the preferred tool for differential protein expression and biomarker discovery.
These experiments areÂ performed on a hybrid quadrupole/time-of-flight tandem mass spectrometer (Synapt G2S)Â coupled with ultra-performance nanoscale capillary liquid chromatographyÂ (LC/ESI/MS/MS)
